  Background Protein kinase B(Akt)plays important roles in regulation of cell growth and survival,but little is known about the AKT3(Protein kinase B γ)function,especially the action of AKT3 on human invasive breast ductal carcinoma(IBDC).Methods Expression of AKT3 was analyzed by immunohistochemistry in 80 cases of IBDC tissues.BT474 IBDC cell migration and invasion were quantified using the Transwell assay,and the cell apoptosis and survival were detected by DNA ladder and cell survival assays.The Wilcoxon Signed Ranks test,Chi-square test,Fishers exact,and independent T test were used for analysis.Results AKT3 was over-expressed in 57 of 80 cases(71.2%)of IBDC tissues,and was associated with higher TNM stage and lymph node metastasis,but not patient age or tumor size.Epidermal growth factor (EGF)increased cell migration,invasion and survival,and decreased cell apoptosis in AKT3 over-expressed BT474 cell,which was inhibited by PI3K/AKT pathway inhibitor wortmannin.Conclusion Over-expressed AKT3 was associated with higher TNM stage and lymph node metastasis in IBDC.In vitro studies indicated over-expressed AKT3 may increase the metastatic and survival ability of IBDC cells.
