Transient receptor potential melastatin 8 (TRPM8) is a member of the melastatin subfamily of TRP channels and well known for its role in sensory neurons as the cold sensor.TRPM8 channels can be activated by cold temperatures and cooling agents such as menthol and icilin. In this study we found that the comeal permeability-enhancing drug (+)-borneol significantly increased tear production in guinea pigs, which is similar to the action of menthol.Using a HEK293 cell line stably expressing TRPM8 channels, we demonstrated that micromolar borneol activated TRPM8 channels by intracellular Ca2+ measurement and patch-clamp recordings.
