The ability to correlate single-cell genetic and protein expression information to cellular phenotypes will provide the kind of detailed insight into human physiology and disease pathways that is not possible to infer from bulk cell analysis.This capability is particularly pertinent to understanding cancer biology,given the highly heterogeneous nature of the disease.This presentation describes a set of single-cell isolation,manipulation,and analysis techniques that we have developed in our lab,including a rare-cell isolation platform we called eDAR,the active manipulation and trapping of single cells in microfluidic systems,the analysis of single-cell gene expression with digital PCR,and the quantification of expressions of proteins at the single-cell level.
