Isomerase is an enzyme mediates conversion of monosaccharides between aldose and ketose,and sub-categorized depending on the reaction mechanisms.To investigate the evolutionary characteristics of two different isomerases,a metallic isomerase (EC 5.3.1.4,arabinose isomerase from Escherichia coli:AI) and a non-metallic isomerase (EC 5.3.1.26,tagatose-6-phosphate isomerase from Staphylicoccus aureus:TPI) were selected as a model.Random mutations were introduced with rote of 2 mutations per 1Kb to the model isomerase genes,and activities of galactose isomerization,a non-natural substrate,were determined.About 50% of AI mutant derivatives showed increase of galactose isomerization activity,while only about 30% of TPI mutant derivatives increased the activity.
