The postsynaptic density (PSD) has been a target for massive proteomics analysis during the past 10 years and recent proteomics analyses have suggested the presence of several or even more than 1,000 species of proteins localized in the PSD.To identify postsynaptic proteins massively,we attempted an identification ofpostsynaptically localized mRNA species,based on the idea that massive identification of them enables efficient identification of the postsynaptic proteins.We isolated and characterized mRNAs associated with PSD prepared from the rat forebrain by random sequencing and gene chip microarray analysis.We found that a large number of mRNA species are associated with the PSD fraction and that mRNAs encoding various postsynaptic proteins,such as channels,receptors,proteins involved in signaling,scaffold and adaptor proteins and cytoskeletal proteins,were highly concentrated in the PSD fraction,whereas those encoding housekeeping proteins,such as enzymes in the glycolytic pathway,were not.
