The transcription factor E2F, the main target of the tumor suppressor RB, plays central roles in activation of growth-promoting genes through physiological inactivation of RB by growth stimulation.E2F can also activate pro-apoptotic and growth-suppressive genes, whose regulation is not well understood.We previously reported that the tumor suppressor ARF gene, an upstream activator of the tumor suppressor p53, and CDK inhibitor p27Kipl gene are activated by deregulated E2F induced by RB-dysfunction, a major oncogenic change, but not by physiological E2F induced by growth stimulation in human normal fibroblasts (HFFs).
