Proper dendritic arborization is essential for establishing normal neuronal circuitry.Here we report a role of geranylgeranyltransferase Ⅰ (GGT),a prenyltransferase which mediates lipid modification of Rho GTPases for their association with the plasma membrane,in dendritic morphogenesis.We found that GGTα level was gradually increased during the period of dendrite growth in vivo and enriched in dendrites.Inhibition or down-regulation of GGT caused a decrease in dendrite length and branches,whereas over-expression of GGT promoted dendrite development of cultured hippocampal neurons.Interestingly,KCl depolarization or treatment with BDNF quickly increased GGT activity,resulting in increased membrane association of Racl.In line with this notion,GGT was physically associated with TrkB,the receptor of BDNF.The effect of KC1 depolarization or BDNF on dendrite development was dependent on GGT,since GGT inhibition attenuated dendrite growth induced by high KCl or BDNF.Two mutated forms of Racl,Rac 1AC and RaclC189S which lost its membrane location characteristic could attenuated GGT-induced dendrite development.Thus,these results identify GGT as an important regulator for activity-or BDNF-induced dendritic morphogenesis.
