星形胶质细胞调节T细胞因子的分泌

  星形胶质细胞的免疫调控功能已受到广泛关注.不同状态(静息/活化)的星形胶质细胞可调节T细胞增殖和凋亡,并影响T细胞因子的分泌.Th1和Th2型细胞因子在CNS炎性疾病的发生发展过程中发挥不同角色,本研究利用IFN-γ刺激活化星形胶质细胞,观察其调节T细胞因子(Th1/Th2)的分泌.本实验采用ELISA的方法检测T细胞分泌的细胞因子IL-2、IFN-γ、IL-4和IL-10,明确Th1型和Th2型细胞因子的分泌,以深入了解星形胶质细胞对T细胞的调控作用.结果表明星形胶质细胞经接触模式可抑制T细胞分泌IL-2、IFN-γ,活化后的星形胶质细胞抑制效应减弱,而IL-4和IL-10的分泌变化不明显,提示星形胶质细胞通过接触培养后,对Th1细胞因子的分泌明显受到抑制,其中活化后的抑制效应降低.采用Transwell模型共培养星形胶质细胞与T细胞,检测Th1/Th2细胞因子的分泌,发现活化前后的星形胶质细胞均能抑制细胞因子IL-2、IFN-γ的分泌,而IL-4和IL-10的分泌也未受影响,提示星形胶质细胞分泌细胞介质对Th1细胞因子的分泌具有抑制作用.
