  Prostaglandin (PG) D2 is one of the most potent sleep-promoting substances.PGD2-induced sleep was inhibited by an antagonist of adenosine A2A receptors (A2AR),suggesting that adenosine A2AR mediates the sleep-promoting effect of PGD2.In the present study,we used A2AR knockout (KO) mice to further explore the involvement of adenosine receptor subtypes in the somnogenic effect of PGD2.An A2AR agonist,CGS21680,at doses of 0.04,0.2,1 and 5 pmol/min increased non-rapid eye movement (NREM) sleep in wild-type (WT) mice after infusion into the lateral ventricle in a dose-dependent manner,but not at all in A2AR KO mice,as compared with the baseline day.In contrast,an agonist of adenosine A1 receptors (A1R),cyclopentyladenosine,did not affect sleep profiles in WT mice at a dose of 1 pmol/min and increased NREM sleep slightly at a dose of 5 pmol/min.
