During the neuronal development, the tremendous but tightly regulated expansion of plasma membrane is required for neurite outgrowth and arborization. However the underlying mechanism which regulates this process remains largely unknown. Here we report a role of Lgll,a mammal homolog of Lethal giant larvae which is involved in secretion pathway of yeast,in neural development.Lgl1 is highly expressed in the rat brain.A large fraction of Lgl1 was associated with membrane. We found that Lgl1 was critical for dendrite outgrowth and arborization, since overexpression of Lgl1 in cultured hippocampal neurons resulted in an increase of dendrite length and branches whereas down-regulation of Lgl1 decreased dendrite growth.
