Alzheimers disease (AD) is the most common progressive neurodegenerative disorder that is believed to be due to the overproduction (or under-removal) and subsequent aggregation of short,hydrophobic Aβ peptides derived from amyloid precursor proteins (APP) by proteolytic cleavage.The two Aβ peptides,Aβ40 and Aβ42,are generated by the BACEl and γ—secretases acting in concert.However,due to multiple substrates of these secretases as well as their low enzymatic specificity of aspartyl protease family,little progress has been achieved in clinical trials of secretase inhibitors/mediators.
