长非编码RNA与miRNA介导的泛癌调控网络

  非编码RNA被认为在基因表达调控与多种人类疾病的发生发展中起到重要作用,但目前人们对多种类型的非编码RNA之间的调控网络知之甚少.我们开发了starBase v2.0 (http://starbase.sysu.edu.cn/)分析平台,通过整合来自39个独立研究产生的111个CLIP-Seq数据来系统地鉴定RNA-RNA以及RNA结合蛋白(RBP)-RNA的相互作用网络.通过分析上百万个RBP结合位点数据,我们鉴定出~10,000个miRNA-IncRNA,~9,000个miRNA-circRNA,~16,000个miRNA-pseudogene,~450,000个miRNA-mRNA以及~285,000个RBP RNA调节关系.我们通过挖掘TCGA数据库中14种癌症的临床与表达数据,从泛癌的角度进一步分析了这些调控网络,并构建了CLIP数据与癌表达数据支持的~200,000 ceRNA对(竞争性内源RNA对).通过这些ceRNA对形成的调控网络,结合13种功能基因注释信息,我们开发了能对IncRNA、miRNA及蛋白基因的功能进行预测的算法.最后我们将上述数据的可视化与分析过程进行了网页实现.我们的研究将对我们理解非编码RNA的功能以及它们介导的调控网络在癌症中的功能起到推进作用.
