It is known that the envelope (env) gene of pathogenic agent type III human T-lymphotropic virus (HTLV-III) of acquired immunodeficiency syndrome (AIDS) defines a first order polypeptide of about 860 amino acids which is glycosylated A precursor of 160,000 in molecular weight (gp160), and further the mature membrane-associated proteins gp120 and gp41. This article describes the expression of the complete env gene by vaccinia virus vectors and provides evidence of the synthesis, glycosylation, processing, and membrane trafficking of env polypeptides. The authors linked a DNA fragment containing the entire env gene of HTLV-III to the downstream of the vaccinia virus promoter to form the env-leader recombinant vaccinia viruses V25 and V26 containing 0 and 70 base pairs (bp), respectively. Both recombinants were thymidine kinase negative and beta-galactosidase positive.
