目的:探讨L5脊神经结扎(SNL)术后7 d和28 d大鼠脊髓背角内胶质细胞数量的改变与意义。方法:3月龄雄性SD大鼠随机分为SNL组和假手术组,分别于术后7 d和28 d取腰5节段脊髓制作石蜡包埋连续切片50张,等距随机抽选2套切片(每套各5张)分别用IBa-1和GFAP的免疫组织化学染色显示小胶质细胞(microglia,MG)和星形胶质细胞(astrocyte,AS)。采用体视学技术-光学体视框估计单位体积脊髓背角内MG或AS的数量(数密度),将其乘以脊髓背角的横断面积和单位长度(1 mm),即可得到1 mm长脊髓背角内MG和AS的数量。结果:与未手术侧相比,SNL组手术侧1 mm长腰5节段脊髓背角内MG的数量在术后7 d和28 d分别显著增加了210%和120%;AS的数量在术后7 d显著增加了62%,术后28 d无明显增加。结论:神经病理性疼痛可能与胶质细胞数量的动态改变相关。
Objective: To investigate the changes and significance of glial cells in dorsal horn of spinal cord in rats after L5 spinal nerve ligation (SNL) at 7 d and 28 d. Methods: Three-month-old male Sprague-Dawley rats were randomly divided into SNL group and sham operation group. Fifty consecutive spinal paraffin sections were obtained on the 5th and 5th day after operation. Two sets of random paraffin-embedded sections Immunohistochemical staining with IBa-1 and GFAP, respectively, showed microglia (MG) and astrocyte (AS). The volume (number density) of the MG or AS in the spinal dorsal horn of a unit volume is estimated using the stereological technique of the optical system - optical box and multiplied by the cross-sectional area and the unit length (1 mm) of the dorsal horn of the spinal cord to obtain 1 mm The number of MG and AS in the long spinal dorsal horn. RESULTS: Compared with the non-surgical side, the number of MG in the 5-d spinal spinal dorsal horn of the operative side of the SNL group increased significantly by 210% and 120% on the 7th and 28th postoperative days, respectively. The number of AS After 7 d increased significantly by 62%, no significant increase after 28 days. Conclusion: Neuropathic pain may be related to the dynamic changes of glial cells.
