For the differential diagnosis of oral leukoplakia, lichen planus, and carcinoma in situ (Cis for short), using traditional histopathological criteria, microscopy methods have become unsuitable. Tissue measurement techniques can be used to obtain an objective differential diagnosis with a limited number of epithelial histological parameters. This article describes the use of optical microscopy tissue measurement methods, trying to eliminate subjective factors to improve the accuracy of differential diagnosis. 10 cases of leukoplakia, 9 cases of lichen planus, and 14 cases of in situ cancer biopsy specimens were fixed with 10% formalin and stained with hematoxylin-eosin.
