NID1(Nidogen-1)是我们在前期卵巢癌分泌/释放蛋白质组研究中筛选并验证到的的候选卵巢癌诊断血浆蛋白标志物,本研究旨在探索NID1在卵巢癌细胞生物学过程中扮演的角色.Transwell迁移和侵袭实验结果表明瞬时敲降内源性NID1蛋白的HEY细胞其迁移和侵袭能力减弱,同时伴随着细胞外形从间皮细胞样向上皮细胞样转变;Western blot和细胞免疫荧光的结果则显示该细胞中上皮细胞标志分子E-cadherin表达上调,间质细胞标志分子N-cadherin表达下调.综上所述,敲降NID1可促进HEY细胞发生间质-上皮转化(Mesenchymal-Epithelial Transition,MET)来减弱其侵袭转移能力.
