IL-25滴鼻引起小鼠气道周围新生血管形成

目的:探讨IL-25对小鼠气道周围新生血管形成的影响及机制方法:建立周期为70天的慢性哮喘小鼠模型,设立生理盐水组;50 μg OVA激发组;IL-25激发组.ELISA法检测bFGF、VEGF水平;免疫组化法观察vWF+血管数目、ERG、EGF、IGF-1、endotholin-1、angiogenin、amphiregulin等指标的变化;体外细胞实验研究IL-25通过作用于人脐静脉内皮细胞(HUVEC)促进血管再生的功能.结果:和OVA激发组类似,IL-25组小鼠气道周围vWF+及ERG+血管数目显著增多,同时血管再生因子bFGF、VEGF、EGF、IGF-1、endothelin-1、angiogenin和amphiregulin表达增加;IL-25能够引起HUVEC增殖,并出芽生长,形成新生的微血管结构.
