The countless possibilities for the precise engineering of proteins via the expanded genetic code in living cells have been hampered by the poor incorporation of unnatural amino acid (UAA),particularly at multiple sites.Here we explored the assignment of amber codon as either a stop or encoding signal or both in bacterial by CRISPR interference (CRISPRi) targeting release factor 1 (RF1),an endogenous protein allowing for termination of translation.
