Objective: Establishing patient-derived subrenal capsule (SRC) xenografls in pancreatic cancer (PC) as a novel pre-clinical model for predicting chemotherapeutic responsiveness.Methods: 24 PC patients tumour xenografted under the renal capsules in NOD/SCID mice.Following 4 weeks xenografts, the mice randomly received either Gemcitabine or saline (control) for further 4-weeks treatment.Gemcitabine responses were determined by: the mean post-graft area between treatment groups, the histological morphology, the inmmunohistochemical (IHC) scores and correlation with clinical outcome.Results: SRC xenograft model was successfully established with a rate of 91.57%.Original and post-grafts tumours demonstrated similar histopathological features.7 were unresponsive to Gemcitabine in mice among 8 patients developed metastasis or deceased within 25 days to 10 months.5 patients whose tumours responded to Gemcitabine in mice, 3 of them have not developed recurrence or metastasis and other 2 patients developed recurrence or metastasis after surgery longer than 12 months.Conclusion: The high xenografls rate of SRC model allows using it as a potential tool to assess an individual patient response to Gemcitabine treatment within 8 weeks after pancreatic tumour resection.
