Zebrafish is an excellent model for circadian studies.Lack of mutants for circadian clock genes,however,has hindered zebrafish circadian research.Here,we used TALEN to generate two lines of null mutants for zebrafish period2(per2).Locomotor experiments showed that the per2 mutant fish display reduced activity amplitude under LD(light-dark)condition and 2-hour phase delay under DD(dark-dark)condition compared with wild types.Real-time quantitative PCR showed up-regulation of bmal1a,cry1ab but down-regulation of per1a,per1b,bmal1b and bmal2 in the per2 mutant fish.In Vitro cell transfection assays showed that Per2 not only acts through E-boxes to repress E-Box-containing genes but also acts through RRE-boxes to activate RRE-box-containing genes.These results showed that Per2 plays dual roles in zebrafish circadian regulation.
