Somatic cell nuclear transfer(SCNT)is a unique approach to reprogram fully differentiated nuclear to totipotent status for the organism cloning.It has been achieved in many species including fish which owing to Tong’s pioneer work about half century ago.However,the success rate is unsatisfied making the application of the technique very limited.Abundance of evidence showed that there are many epigenetic defects in NT embryos.Epigenetic modifications play important roles both in the development of wild type and nuclear transfer embryos.5-methylcytosine(5mC)is one of the well known epigenetic modifications which is called “the fifth base” for its considerable function in gene regulation.The primary data in other animal models suggested the disorder patterning of 5mC in NT embryos.To further disclose the circumstance of 5mC during NT zebrafish embryos development,we generated a series of zebrafish NT embryos with different donors and explored their single-base resolution DNA methylomes.By compare the methylomes between NT and wild type embryos in detail,it might provide us the full view of 5mC during early development of zebrafish NT embryos lighting the foreground for the mechanic study of zebrafish SCNT reprogramming.
