There are six cry genes in zebrafish.Among them,cry1a acts as a repressor in the circadian clock feedback loop. Here we used TALEN to generate cry1a mutants. We determined that the mutations significantly affect transcription of cry1a.The locomotor activity assays showed that cry1a mutants exhibit hyperactivity compared with wild types under LD(light-dark)condition.Real-time PCR showed that the expression patterns of a number of circadian clock genes are significantly altered in the cry1a mutants. These results showed that Cry1a is essential for zebrafish circadian regulation.
