Parkinson's disease (PD) is one of the most prevalent neurodegenerative diseases characterized by accumulation of α-synuclein (α-syn).Geniposide (Gde) is a major iridoid glycoside isolated from the fruit of Gardenia jasminoides Ellis., which has been widely used in traditional Chinese medicine.In this study, the novel function of Gde as a neuronal autophagy inducer was reported.Gde induced autophagy in different neuronal cell lines, including N2a,PC12 and SH-SY5Y cells.Moreover, Gde promoted autophagy in Drosophila fat body and in the brain of ICR mice.The neuroprotective effect of Gde was evaluated on Drosophila PD model overexpressing wild-type α-synuclein (WT-αsyn).Gde down-regulated the expression level of WT-αsyn in the brains of the flies.The motor function of the flies receiving Gde was improved.The loss of dopaminergic neurons was also ameliorated by Gde.Collectively, we have identified Gde as a neuronal autophagy inducer, which can promote the clearance of WT-αsyn and protect neurons.
