In S.cerevisiae the Cytoplasm to Vacuole Targeting (Cvt) pathway is a type of selective autophagy-related pathway, through which hydrolytic enzymes, like the aminopeptidase Ape1,are sequestered into double-membrane vesicles and transported to the vacuole.Our studies aim to structurally understand the mechanisms that selectively incorporate the Ape1 cargo into the Cvt vesicles.Since the Cvt pathway shares homologies with selective autophagy in higher eukaryotes, these results will elucidate structurally a prototype of selective autophagy from the point of view of the cargo-recognition, cargo-uptake and cargo-packaging.To this end, we have used the Ape1 enzyme, both in its premature (preApe1) and mature (mApe1) forms, as our cargo model.We have determined the low-resolution three-dimensional structure of mApe1 using electron microscopy and we have set up a system that will allow us to visualize ex-vivo the Cvt vesicles with electron cryo-microscopy techniques.
