Successful establishment of metastasis by neoplasms depends on the overcoming of organ specific barriers as well as on the microenvironment dynamics on-site.It has been suggested that tumour cells in the primary lesion can create a pre-metastatic niche at the target organ to promote tumour cell adhesion and colonization.The precise mechanisms by which this could be accomplished are not well characterised.It is possible that tumour cells are able to prime the endothelium of distant organs to enhance adhesion and facilitate the extravasation of circulating tumour cells.
