麦角胺是一种天然存在的生物碱,用于治疗周期性偏头痛。口服或直肠给药均有作用。在体内代谢很快,口服用药后经受较大的“首过效应”。对麦角胺的药动力学和生物利用度研究的困难之一,是它按小剂量给药,在血液和其他体液中难以检测和分析。随着质谱分析技术的发展,已能在微微克水平上分析麦角胺和其它药物的血样。这使得对麦角胺的药动学研究成了可能。研究中有24名受试者参加,每人在不同时间内口服或直肠给予麦角胺2mg。本项研究是双向随机交差法,接受两次剂量至少间隔一周。受试者用药前10小时和用药后4小时禁食;直肠用药前1小时
Ergotamine is a naturally occurring alkaloid used to treat cyclical migraine. Oral or rectal administration has a role. Metabolism in the body quickly, after oral administration to undergo a larger “first pass effect.” One of the difficulties in studying the pharmacokinetics and bioavailability of ergotamine is its low dose administration, which is difficult to detect and analyze in blood and other body fluids. With the development of mass spectrometry techniques, blood samples of ergotamine and other drugs have been analyzed at the picogram level. This makes the pharmacokinetics of ergotamine possible. Twenty-four subjects were enrolled in the study, and 2 mg of ergotamine was administered orally or rectally to each person at different times. This study is a two-way randomized crossover method, receiving two doses at least one week apart. Subjects were fasted 10 hours before and 4 hours after treatment; 1 hour before rectal administration
