本文报告筛选人轮状病毒阳性粪便样品的方法和分离纯化有模板活性的HRV ss RNAs,然后在麦胚无细胞蛋白合成系统中进行翻译。结果每毫克HRV ss RNA能合成2~3.5×10~6cpmTCA不溶的HRV各类多肽;经凝胶电泳分析,在1.1×10~6cpm的各种多肽中,相当于McCrae等人报道的VP_1—VP_4四种多肽位置的放射性为5.4×10~5cpm,相当于VP_5—VP_(12)八类多肽位置的放射性为5.3×10~5cpm左右,其数量的分布趋势也相似。
This article reports a method for screening human rotavirus positive stool samples and isolating and purifying template-competent HRV ss RNAs, which are then translated in a wheat germ cell-free protein synthesis system. Results HRV ss RNA per milligram was able to synthesize 2 ~ 3.5 × 10 ~ 6 cpm TCA-insoluble HRV polypeptides. The results of gel electrophoresis showed that VP1-VP_4 The radioactivity of these four peptides was 5.4 × 10 ~ 5 cpm, which corresponded to about 5.3 × 10 ~ 5 cpm radioactivity of the eight peptide positions of VP_5-VP_ (12). The distribution trend of the number of these peptides was also similar.
