本文报告HSF 79-2二倍体成纤维细胞系,于体外保持较长时期生长增殖的特性,以及其形成组织样结构的初步结果。 HSF79-2细胞系来自17岁女性卵巢癌患者的腹部正常皮肤。原代采用组织块培养法,培养液系McCoy 5 A补加10~25%小牛血清。细胞系以1:2分种率传代、46~54代终止。此系细胞可停止传代,保持原瓶换液而长期生长(观察16个月),并增殖成厚膜。经组织学检查为结缔组织样结构。非传代瓶中保留细胞分别于3,6,12个月后重新传代,细胞的生长特性与二倍体性未变。其总的生存代数与连续传代细胞大致相等。上述特点,可能为二倍体成纤维细胞所共有,可用于保存某些细胞系,为研究细胞的运动、分化提供模型。
This article reports the HSF 79-2 diploid fibroblast cell line in vitro to maintain a longer period of growth and proliferation characteristics, as well as its initial formation of tissue-like structure results. The HSF79-2 cell line is derived from normal abdominal skin of a 17 year old female ovarian cancer patient. The primary use of tissue culture, culture medium McCoy 5 A supplemented with 10 ~ 25% calf serum. Cell lines were passaged at 1: 2 seed rate and terminated at 46-54. This line of cells can stop the passage, to maintain the original bottle replacement fluid and long-term growth (observed 16 months), and proliferate into thick film. Histologically examined as connective tissue-like structure. The cells in non-subculture bottles were resubmitted after 3, 6 and 12 months, respectively. The growth characteristics and diploidity of the cells were unchanged. The overall survival algebra is approximately the same as serial passage cells. These features, may be shared by diploid fibroblasts, can be used to save some cell lines, in order to study the cell movement and differentiation model.
