本文混合应用我室制备的三株抗人肺癌单克隆抗体F—19、SIA—2,N—35,对100例各型肺癌石腊切片标本进行了免疫组化(ABC法)检测,总阳性率91%。其中肺鳞癌93%,肺腺癌86%,肺泡细胞癌83%,肺混合癌(鳞+腺)100%,肺未分化癌91%,较之单个抗体检测大大提高了阳性率。尤其对肺未分化癌(小细胞癌达100%)。避免了单一抗体检测阴性而丢失的阳性病例,并对检测中出现的核抗原作了初探。采用抗原竞争抑制试验证明三株抗体分别针对不同的抗原决定簇,用免疫封闭试验排外CEA抗体。
In this paper, three anti-human lung cancer monoclonal antibodies F-19, SIA-2, N-35 prepared by our laboratory were mixedly applied. Immunohistochemical (ABC) detection was performed on 100 samples of various types of lung cancer paraffin sections. The total positive was positive. The rate is 91%. Among them, 93% of lung squamous cell carcinoma, 86% of lung adenocarcinoma, 83% of alveolar cell carcinoma, 100% of mixed lung adenocarcinoma (squamous + gland), and 91% of lung undifferentiated carcinoma, which greatly increased the positive rate compared with single antibody detection. Especially for lung undifferentiated carcinoma (small cell carcinoma up to 100%). The detection of negative nuclear antigens in the detection of single-antibody-negative positive cases was avoided. The antigen-competitive inhibition assay was used to prove that the three antibodies were directed against different antigenic determinants and that the CEA antibodies were excluded using an immunostaining assay.
