目的:探讨前列宁胶囊(QLNC)治疗良性前列腺增生(BPH)过程中对炎症因子IL-10、TNF-α表达的影响。方法:将成年SD大鼠随机分为正常组,BPH模型组,保列治治疗组(0.5mg.kg-1.d-1),前列宁高、中、低治疗组(2.25g.kg-1.d-1、4.5g.kg-1.d-1、9g.kg-1.d-1),每组10只,采用去势后皮下注射丙酸睾酮法建立大鼠BPH模型(5mg.kg-1.d-1)。28天后观察各组大鼠前列腺湿重和体积、指数及病理形态学变化,RT-PCR法检测大鼠前列腺组织中IL-10、TNF-α表达情况。结果:QLNC组大鼠前列腺体积、指数较模型组明显减小,病理变化较模型组明显改善;RT-PCR显示QLNC能增强大鼠前列腺组织中IL-10表达,降低TNF-α表达。结论:QLNC能明显抑制大鼠前列腺组织增生,降低炎症因子TNF-α在前列腺组织中的表达,提高IL-10在前列腺组织中的表达,表明QLNC通过抑制炎症因子表达可能是治疗BPH的机制之一。
Objective: To investigate the effect of Qianliening Capsule (QLNC) on the expression of inflammatory cytokines IL-10 and TNF-α during the treatment of benign prostatic hyperplasia (BPH). Methods: Adult SD rats were randomly divided into normal group, BPH model group, Baoreng treatment group (0.5mg.kg-1.d-1), Qianliening high, middle and low treatment group (2.25g.kg- 1.d-1, 4.5g.kg-1.d-1, 9g.kg-1.d-1), 10 rats in each group. BPH model was established by subcutaneous injection of testosterone propionate kg-1.d-1). After 28 days, the wet weight, volume, index and histopathological changes of prostatic tissue in each group were observed. The expression of IL-10 and TNF-α in prostatic tissue was detected by RT-PCR. Results: Compared with the model group, the volume and index of prostate in QLNC group were significantly decreased and the pathological changes were significantly improved compared with the model group. The results of RT-PCR showed that QLNC enhanced the expression of IL-10 in prostate tissue and decreased the expression of TNF-α. Conclusion: QLNC can significantly inhibit the proliferation of rat prostate tissue, decrease the expression of inflammatory cytokine TNF-α in prostate tissue and increase the expression of IL-10 in prostate tissue, indicating that QLNC may be the mechanism of BPH treatment by inhibiting the expression of inflammatory cytokines one.
