AIM:To study the anti-neoplastic effect of Haimiding andits mechanisms of action.METHODS:Experiments using MTT and colony formationwere carried out to study the in vitro anti-neoplastic actionof Haimiding,its in vivo anti-neoplastic action was studiedby observing its effect on the weight of tumors in FC miceand S_(180),H_(22) tumor bearing mice,as well as their life spans.The effect of Haimiding on cell apoptosis and differentstages of cell cycles in human gastric carcinoma cells werestudied by flow cytometry.Its effect on [Ca~(2+)]_i of humangastric carcinoma cells and the source of Ca~(2+) during thechange of [Ca~(2+)]_i were observed by confocal laser scanningtechnique.RESULTS:Haimiding showed a definite cytotoxicity to 8human tumor cell lines,which was most prominent againstBGC-823,E_(ca-109) and HCT-8 tumor cells.It also exhibited anobvious inhibition on colony formation of the above tumorcell lines,which was most prominent in E_(ca-109) tumor cells.Itshowed obvious inhibition on the growth of tumor in FCmice and S_(180) bearing mice as well as prolonged the lifespan of H_(22) bearing mice.It was able to induce apoptosisand elevate intracellular [Ca~(2+)]_i concentration of tumor cells.The source of Ca~(2+) came from both extracellular Ca~(2+) influxand intracellular Ca~(2+) release.CONCLUSION:Haimiding is composed of a TCM preparationand 5-flurouracil.Its anti-neoplastic potency is highlyenhanced by synergism as compared with either one of itscomponents.Its mechanisms of anti-neoplastic action canbe attributed to its action to initiate apoptosis of tumorcells by opening the membrane calcium channel andinducing intracellular Ca~(2+) release to elevate [Ca~(2+)]_i of thetumor cells.
