目的:定量检测转化生长因子β受体-1(TransformingGrowthFactor-βreceptortype1,TβR1)和转化生长因子β受体-2(TransformingGrowthFactor-βreceptortype2,TβR2)编码基因在糖尿病大鼠视网膜中不同时相点的动态表达水平,以探讨转化生长因子β及转化生长因子β受体在糖尿病视网膜病变(DR)中的作用。方法:选择健康成年SD大鼠28只,随机分为正常对照组(CON)和糖尿病组(DM)。利用链尿佐菌素(STZ)诱导大鼠糖尿病视网膜病变模型,制备RNA并逆转录,实时荧光定量PCR技术分析TβR1和TβR2在不同时相点相对于内参照基因18s的相对mRNA含量。结果:TβR1在4,12wk时相对于18s的mRNA含量分别为0.000493±0.000133和0.000608±0.000232。TβR2在4,12wk时相对于18s的mRNA含量分别为0.000166±0.000057和0.000113±0.000049。TβR1基因表达水平随着病变进展呈上升趋势,TβR2在4wk时表达量较正常增加,到12wk时表达量又减少,逐渐恢复到正常水平。结论:糖尿病大鼠视网膜中TβR1和TβR2基因表达水平变化可能影响TGF-β信号的转导过程,TGF-β及其受体介导的信号转导通路在糖尿病视网膜病变中具有重要作用。
OBJECTIVE: To quantitatively detect the dynamic expression of Transforming Growth Factase-β Receptorypeptide 1 (TβR1) and Transforming Growth Factase-β Receptorypeptide 2 (TβR2) at different time points in the retina of diabetic rats Level to explore the role of TGF-β and TGF-β in diabetic retinopathy (DR). Methods: Twenty-eight healthy adult SD rats were randomly divided into normal control group (CON) and diabetic group (DM). The model of diabetic retinopathy was induced by streptozotocin (STZ), RNA was prepared and reverse transcribed. The relative mRNA content of TβR1 and TβR2 at different time points relative to the 18s reference gene was analyzed by real-time fluorescence quantitative PCR. Results: The mRNA levels of TβR1 at 4 and 12wk relative to 18s were 0.000493 ± 0.000133 and 0.000608 ± 0.000232, respectively. The mRNA levels of TβR2 relative to 18s at 4 and 12wk were 0.000166 ± 0.000057 and 0.000113 ± 0.000049, respectively. The expression level of TβR1 increased with the progression of the disease. The expression level of TβR2 increased more than normal at 4wk, and then decreased at 12wk and returned to normal. CONCLUSION: The changes of TβR1 and TβR2 gene expression in the retina of diabetic rats may affect the transduction of TGF-β signaling. TGF-β and its receptor-mediated signal transduction pathway play an important role in diabetic retinopathy.
