目的 探讨缺血时细胞外不同Ca~(2+)浓度对神经细胞坏死程度的影响。方法 使用体外培养 12—14天大鼠大脑皮质神经细胞,随机分为正常对照组(8例);单纯缺血组(8例);较高 Ca~(2+)浓度组(8例)高Ca~(2+)浓度组(8例)。检测各组神经细胞内总钙(TCa~(2+)),游离钙(Ca~(2+)i)及细胞外液中乳酸脱氢酶(LDH)浓度变化。结果 单纯缺血组,较高Ca~(2+)浓度组,高Ca~(2+)浓度组细胞内 TCa~(2+)。Ca~(2+)i及外液中LDH 均高于正常对照组(P<0.05),同时高Ca~(2+)浓度组,较高Ca~(2+)浓度组细胞内TCa~(2+)、Ca~(2+)i及外液LDH 浓度均高于缺血组。结论 脑缺血时,随着细胞外液中Ca~(2+)浓度增高,神经细胞内钙沉积增加,从而加速神经细胞死亡。
Objective To investigate the effect of extracellular Ca ~ (2+) concentration on neuronal necrosis during ischemia. Methods Cortical neurons cultured in vitro for 12-14 days were randomly divided into normal control group (8 cases), ischemia group (8 cases) and high Ca 2+ concentration group (8 cases) Ca ~ (2+) concentration group (8 cases). The concentrations of total calcium (TCa 2+), free calcium (Ca 2+) i and extracellular LDH in nerve cells of each group were measured. Results TCa ~ (2+) in the group of pure ischemia, high concentration of Ca ~ (2+), high concentration of Ca ~ (2+) Ca 2+ and LDH in the extracorporeal fluid were higher than those in the normal control group (P <0.05). At the same time, the intracellular TCa ~ (superscript +) in high Ca ~ (2+) and Ca ~ (2+) 2 +), Ca ~ (2+) i and LDH in peripheral blood were higher than those in ischemic group. Conclusions Cerebral ischemia, with increasing concentration of Ca ~ (2+) in extracellular fluid, increases intracellular calcium deposition and accelerates neuronal cell death.
