该文目的为研究LIN28基因对人胰腺癌细胞增殖和迁移的影响,揭示其在胰腺癌发生发展中的作用。实验发现,胰腺癌细胞中LIN28的表达模式与细胞增殖和迁移能力有关,将干扰质粒LIN28 sh RNA转染入LIN28高表达的PANC1细胞,将过表达载体3×Flag-LIN28转染入低表达的SW1990细胞中,采用RT-PCR和Western blot实验验证基因沉默或过表达的效果,并运用CCK-8法分析细胞的增殖能力并绘制细胞生长曲线,Transwell实验检测细胞迁移率。结果显示,下调LIN28后,PANC1细胞中LIN28基因的表达量明显降低,细胞增殖减慢,迁移减少;而上调LIN28后,SW1990细胞中LIN28基因的表达量明显增高,细胞增殖加快,迁移增加。由此说明,沉默LIN28基因可以抑制PANC1细胞增殖和迁移,过表达LIN28基因可以促进SW1990细胞增殖和迁移,这为阐明LIN28在胰腺癌演化过程中的机制奠定了实验基础。
The purpose of this paper is to investigate the effect of LIN28 gene on the proliferation and migration of human pancreatic cancer cells and to reveal its role in the development of pancreatic cancer. The experimental results showed that the expression pattern of LIN28 in pancreatic cancer cells was related to the cell proliferation and migration ability, and the interference plasmid LIN28 sh RNA was transfected into LINANC-highly expressed PANC1 cells. The overexpression vector 3 × Flag-LIN28 was transfected into the low expression In SW1990 cells, the effect of gene silencing or overexpression was verified by RT-PCR and Western blot. The cell proliferation was analyzed by CCK-8 and the cell growth curve was drawn. Transwell assay was used to detect the cell migration rate. The results showed that after the down-regulation of LIN28, the expression of LIN28 gene in PANC1 cells was significantly decreased, the cell proliferation was slowed down and the migration was reduced. However, the up-regulation of LIN28 significantly increased the expression of LIN28 gene in SW1990 cells. Thus, silencing LIN28 gene can inhibit the proliferation and migration of PANC1 cells, overexpression of LIN28 gene can promote the proliferation and migration of SW1990 cells, which laid the experimental foundation for clarifying the mechanism of LIN28 in the process of pancreatic cancer evolution.
