据报导,抗癌光敏剂血卟啉衍生物(HPD),经光照后产生的单线态氧对癌细胞的杀伤作用,在早期主要表现在细胞膜上。另有报导,细胞类脂相变的异常与细胞的恶变有密切的联系。因此,研究HPD对膜相变的影响,对探讨HPD的药理作用有一定的意义。有人用顺磁共振(ESR)技术研究了光照HPD对脂质体膜和对小鼠腹水肝癌细胞膜的作用,并得到了初步结果。为了多途径探讨这一问题,本实验用冰冻断裂电子显微术,研究了光照HPD对二豆蔻酰磷脂酰胆碱(DMPC)脂质体相变的影响。
It has been reported that the anticancer photosensitizer hematoporphyrin derivative (HPD), after irradiation by the singlet oxygen on the killing effect of cancer cells, mainly in the early manifestations of the cell membrane. Another report, abnormal cell lipid phase transition and malignant cells are closely linked. Therefore, the study of HPD on the membrane phase transition, to explore the pharmacological effects of HPD have some significance. Some people use the paramagnetic resonance (ESR) technology to study the illumination HPD on the liposome membrane and mouse ascites liver cancer cell membrane, and obtained preliminary results. In order to explore this issue in many ways, the effect of illumination HPD on the phase transition of liposomes of DMPC was studied by means of freeze-fracture electron microscopy.
