以EMS处理大麦品种花30的游离小孢子,结合低氮胁迫培养获得再生植株,其自交一代的加倍单倍体(DH)株系,在正常施肥和低氮胁迫两种施肥条件下对DH株系进行鉴定,选择株高、单株有效穗和单株产量3个农艺性状作为鉴定指标,并与原始亲本花30进行比较。结果表明:在大田正常施肥和低氮胁迫条件下,DH株系中株高、有效穗和单株产量3个性状出现了明显的正向变异,在低氮胁迫下的表现更为明显。结论:利用游离小孢子的EMS处理结合低氮胁迫筛选培养,可以获取耐低氮性提高的纯合变异体。
Free microspores from barley variety Hua30 were treated with EMS, and regenerated plants were obtained by combining with low nitrogen stress. The doubled haploid (DH) lines were selfed and fertilized under normal and low nitrogen stress conditions for DH The strains were identified, and three agronomic traits, including plant height, effective panicle per plant and yield per plant, were selected as the identification indexes and compared with the original parents 30. The results showed that under the condition of normal fertilization and low nitrogen stress in the field, the three traits of plant height, effective panicle and single plant yield in DH strain showed a significant positive variation, especially under low nitrogen stress. Conclusion: The homozygous mutant with low nitrogen tolerance can be obtained by EMS treatment of free microspores combined with low nitrogen stress screening culture.
