目的建立高效液相色谱法(HPLC)测定灵芝孢子油中1,2-二油酸-3-棕榈酸甘油酯含量的方法。方法采用HPLC法,色谱柱:Diamonsil C18(250 mm×4.6 mm,5μm)柱,流动相:乙腈-异丙醇(体积比为51∶49),流速:1.0 mL·min-1,ELSD漂移管温度:40℃,载气流速:2.4 L·min-1。结果 1,2-二油酸-3-棕榈酸甘油酯在2.7~27.0μg范围内线性关系良好(r=0.9996),平均回收率为99.3%,RSD值为2.3%。结论该方法简便、准确、分离度好,可用于灵芝孢子油中1,2-二油酸-3-棕榈酸甘油酯的含量测定。
Objective To establish a HPLC method for the determination of 1,2-dioleate-3-palmitic acid glyceride in Ganoderma spore oil. Methods The HPLC method was applied on a Diamonsil C18 column (250 mm × 4.6 mm, 5 μm). The mobile phase consisted of acetonitrile-isopropanol (volume ratio 51:49), flow rate 1.0 mL · min -1, ELSD drift tube Temperature: 40 ° C, carrier gas flow rate: 2.4 L · min-1. Results The linear range of 2.7 ~ 27.0 μg was obtained with a good linearity (r = 0.9996). The average recovery was 99.3% and the RSD was 2.3%. Conclusion The method is simple, accurate and has good resolution. It can be used to determine the content of 1,2-dioleoic acid-3-palmitic acid glycerin in Ganoderma spore oil.
