研究载脂蛋白E(ApoE)基因多态性与Ⅱ型糖尿病肾病的关系。方法以载脂蛋白E基因为候选基因,以相互之间无亲缘关系的56例Ⅱ型糖尿病未合并肾病的患者、58例Ⅱ型糖尿病肾病患者和76例正常对照者为对象,应用聚合酶链反应(PCR)扩增从白细胞中提取的载脂蛋白E基因DNA片段,以限制性内切酶HhaⅠ消化,经限制性片段长度多态性(RFLP)分析,区分6种ApoE的基因表型:E2/2、E2/3、E3/3、E3/4、E4/4。结果①3组研究对象的ε2、ε3、ε4等位基因分布频率分别为36%、839%、115%,146%、741%、113%和57%、831%、112%。Ⅱ型糖尿病肾病患者其ε2等位基因频率(146%)显著高于Ⅱ型糖尿病非肾病患者(36%)(P<0005,χ2=837)和正常对照者(57%)(0025<P<005,χ2=428),3组中ε3和ε4等位基因频率差异无显著性(P>005)。②通过比较Ⅱ型糖尿病患者不同的ApoE基因型和血脂的关系发现,ApoE基因多态性和血甘油三酯(TG)、总胆固醇(TC)和低密度脂蛋白胆固醇(LDLC)水平相关,携带ε2等位基因的个体TG水平?
To study the relationship between apolipoprotein E (ApoE) gene polymorphism and type 2 diabetic nephropathy. Methods The apolipoprotein E gene was used as a candidate gene in 56 patients with type 2 diabetes without renal disease, 58 patients with type 2 diabetic nephropathy and 76 normal controls, which were unrelated to each other.
