在体外实验条件下,用~3H-TdR掺入法,观察了钙通道拮抗剂尼卡地平对正常人免疫细胞功能的影响。结果表明,尼卡地平以浓度依赖的方式抑制PHA刺激的外周血T淋巴细胞的增殖及SPA刺激的外周血B淋巴细胞的增殖,IC_(50)分别是:小于10~(-6)mol及8×10~(-5)mol。尼卡地平对外周血白细胞吞噬白色葡萄球菌的影响,显示随着药物浓度降低,抑制作用增强的趋势。上述结果说明,尼卡地平对人的免疫细胞功能具有抑制作用。这和尼卡地平在动物的整体实验结果是一致的。
Effects of Nicardipine, a calcium channel antagonist, on the function of immune cells in normal human beings were observed under ~ 3H-TdR incorporation in vitro. The results showed that nicardipine could inhibit the proliferation of peripheral blood T lymphocytes stimulated by PHA and the proliferation of peripheral blood B lymphocytes stimulated by SPA in a concentration-dependent manner. IC 50 were less than 10 -6 mol and 8 × 10 ~ (-5) mol. The effect of nicardipine on phagocytosis of peripheral white blood cells by Staphylococcus aureus showed that the inhibitory effect was enhanced with the decrease of drug concentration. The above results suggest that nicardipine has an inhibitory effect on human immune cell function. This is consistent with the overall experimental results of nicardipine in animals.
