目的探讨米非司酮(MIF)对宫颈癌裸鼠移植瘤放疗敏感性的影响及其作用机制。方法建立人宫颈癌裸鼠移植瘤模型,筛选32只荷瘤裸鼠,随机分为对照组、放疗组、MIF组、MIF+放疗组4组,每组8只。定期测肿瘤体积,绘制肿瘤生长曲线;处死裸鼠,取瘤体组织,TUENL法检测肿瘤组织细胞凋亡;Western blotting检测各组NF-κBp65蛋白表达水平并进行定位、半定量分析。结果与其他各组相比,MIF+放疗组的裸鼠肿瘤体积下降最明显(P<0.05),细胞凋亡指数最高,肿瘤组织中NF-κB p65蛋白表达水平较单纯放疗组下降明显,差异具有统计学意义(P<0.05)。结论 MIF可通过抑制NF-κB活性,促进细胞凋亡,增强人宫颈癌裸鼠移植瘤的放疗敏感性。
Objective To investigate the effects of mifepristone (MIF) on radiosensitivity of cervical cancer xenografts in nude mice and its mechanism. Methods Nude mice bearing cervical cancer were established. 32 nude mice bearing tumor were randomly divided into control group, radiotherapy group, MIF group and MIF + radiotherapy group, with 8 mice in each group. The tumor volume was measured regularly, and the tumor growth curve was drawn. The nude mice were sacrificed and the tumor tissues were removed. The apoptosis of tumor cells was detected by TUENL assay. The expression of NF-κBp65 protein was detected by Western blotting and semi-quantitative analysis. Results Compared with other groups, the tumor volume of MIF + radiotherapy group was the most significant (P <0.05), and the apoptosis index was the highest. The expression of NF-κB p65 protein in tumor tissue was significantly lower than that of radiotherapy alone group Statistical significance (P <0.05). Conclusion MIF can enhance the apoptosis of human uterine cervix carcinoma xenografts by inhibiting the activity of NF-κB, and enhance the radiosensitivity of human cervical cancer xenografts.
