目的研究前列腺癌细胞系中STAT3活性与顺铂敏感性之间的关系。方法通过免疫细胞化学和蛋白质印迹法(Western blotting)检测3种常见前列腺癌细胞系LNCaP、PC3、DU145基础STAT3的活性;选取激素非依赖性细胞系PC3、DU145,分别加入2 ng/ml、20 ng/ml、200 ng/ml、2μg/ml、20μg/ml顺铂检测细胞增殖抑制情况,蛋白质印迹法检测低浓度顺铂作用DU145后STAT3的活性变化。结果激素非依赖性前列腺癌细胞系PC3、DU145中STAT3的活性较激素依赖性细胞系LNCaP中的高,且STAT3活性较低的PC3细胞较DU145细胞对顺铂更敏感,低浓度顺铂长时间作用DU145后可引起STAT3活性上调。结论 STAT3可能参与调节前列腺癌细胞对顺铂的敏感性。
Objective To investigate the relationship between STAT3 activity and cisplatin sensitivity in prostate cancer cell lines. Methods The basic STAT3 activities of three kinds of prostate cancer cell lines, LNCaP, PC3 and DU145, were detected by immunocytochemistry and Western blotting. The hormone-independent cell lines PC3 and DU145 were selected and added with 2 ng / ml, 20 The inhibitory effect of cisplatin on the proliferation of DU145 cells treated with low concentration cisplatin was detected by Western blotting. Results The STAT3 activity in hormone-independent prostate cancer cell lines PC3 and DU145 was higher than that in hormone-dependent cell line LNCaP, and PC3 cells with lower STAT3 activity were more sensitive than cisplatin to DU145 cells.
