目的研究CyclinD1和P73在口腔粘膜鳞状细胞癌中应用奥铂联合替加氟化疗前后的表达及临床意义。方法选取30例口腔粘膜鳞状细胞癌患者的癌组织为实验组,10例口腔粘膜良性肿瘤患者的正常黏膜为对照组。将癌组织和正常粘膜组织分为三组,即A组(化疗前)、B组(化疗后)和C组(正常粘膜)。应用免疫组织化学SP法和流式细胞术,检测CyclinD1和P73在各组中的表达。结果①免疫组化:CyclinD1在化疗前口腔粘膜鳞状细胞癌中阳性率为91.29±3.10%,化疗后为71.78±4.46%,正常黏膜组织阳性率为51.32±2.29%;P73在化疗前口腔粘膜鳞状细胞癌中阳性率为88.97±4.2%,化疗后为72.14±11.6%,正常黏膜组织阳性率为53.49±5.01%。②流式细胞术:CyclinD1在化疗前口腔粘膜鳞状细胞癌中的表达为1.89±0.10,化疗后为1.35±0.15,正常口腔粘膜细胞为1.0±0.04。各组之间比较有显著性差异(P<0.01)。P73在化疗前为1.75±0.18,化疗后为1.29±0.15,正常粘膜细胞为0.99±0.04。各组之间比较有显著性差异(P<0.01)。结论 CyclinD1和P73可能是口腔黏膜癌变的早期事件,可作为癌变监测和临床疗效观察的辅助指标。
Objective To investigate the expression and clinical significance of CyclinD1 and P73 in oral mucosal squamous cell carcinoma before and after the combination of Octreotide and Tegafur. Methods Tumor tissues from 30 patients with oral mucosal squamous cell carcinoma were selected as experimental group and 10 normal mucosa as control group. The cancer tissue and normal mucosa were divided into three groups, namely, group A (before chemotherapy), group B (after chemotherapy) and group C (normal mucosa). Immunohistochemical SP method and flow cytometry were used to detect the expression of CyclinD1 and P73 in each group. Results ① Immunohistochemistry: The positive rate of CyclinD1 in oral mucosal squamous cell carcinoma was 91.29 ± 3.10% before chemotherapy, 71.78 ± 4.46% after chemotherapy and 51.32 ± 2.29% in normal mucosa. The positive rate of P73 in oral mucosa The positive rate of squamous cell carcinoma was 88.97 ± 4.2%, 72.14 ± 11.6% after chemotherapy, and the positive rate of normal mucosa was 53.49 ± 5.01%. ② Flow cytometry: The expression of CyclinD1 in squamous cell carcinoma of oral mucosa before chemotherapy was 1.89 ± 0.10, 1.35 ± 0.15 after chemotherapy, and 1.0 ± 0.04 in normal oral mucosa. There was significant difference between each group (P <0.01). P73 was 1.75 ± 0.18 before chemotherapy, 1.29 ± 0.15 after chemotherapy, and 0.99 ± 0.04 for normal mucosal cells. There was significant difference between each group (P <0.01). Conclusion CyclinD1 and P73 may be early events of oral mucosal carcinogenesis, which can be used as auxiliary indicators for monitoring of cancer and clinical curative effect.
