脱氧5-氟尿苷(5′-DFUR)是1979年由Cook等人合成的5-FU诱导剂,磷酸嘧啶核苷在肿瘤组织内具有高活性,能使5′-DFUR转化成5-FU。5′-DFUR对胃癌、直肠癌、乳腺癌有效,用于头颈部恶性肿瘤较少。本文作者以7例口腔癌患者为对象,其中原发癌4例、局部复发及复发转移癌3例,术前1周口服5′-DFUR400mg,然后行化疗,以CDDP和PEP方案为主,放疗4例,再行手术切除肿瘤,测定切除肿瘤组织及相连接正常组织5-FU浓度,部分患者测定血中5-FU浓度,研究了5′-DFUR的安全性及有效性。
Deoxy-5-fluorouridine (5’-DFUR) is a 5-FU inducer synthesized by Cook et al. in 1979. Pyrimidine nucleoside has high activity in tumor tissues and can convert 5’-DFUR to 5-FU. . 5′-DFUR is effective for gastric cancer, rectal cancer, and breast cancer, and is used for less head and neck malignancies. The authors of this study were 7 patients with oral cancer, including 4 cases of primary cancer, 3 cases of local recurrence and recurrence of metastatic cancer, oral administration of 5’-DFUR 400mg 1 week before surgery, followed by chemotherapy, mainly CDDP and PEP regimens, radiotherapy In 4 patients, the tumors were resected and the concentration of 5-FU in the removed tumor tissues and normal tissues was determined. Some patients were tested for the concentration of 5-FU in blood, and the safety and efficacy of 5′-DFUR were studied.
