人T淋巴细胞用低剂量的X射线处理后,可以抵抗大剂量X射线引起的细胞突变反应.研究中使用的T细胞是从外周血分离出来的,进行体外培养.培养液RPMI 1640含有2mmol的谷氨酰胺、100U/ml青霉素、100μg/ml链霉素和20%的HL-1培养液.细胞浓度为10~5/ml,用PHA刺激细胞有丝分裂.在PHA作用24小时后实验组给以1cGy的X线照射,在PHA作用24小时后,用300cGy的X射线照射,细胞培养液加入20%的T细胞生长因子(淋巴因子作用于K细胞后的上清液)培养6~8天,使HPRT~-突
Treatment of human T lymphocytes with low-dose X-rays can protect against large cell X-ray induced cell mutagenesis. The T cells used in the study were isolated from peripheral blood and cultured in vitro. The culture RPMI 1640 contained 2 mmol of Glutamine, 100 U/ml penicillin, 100 μg/ml streptomycin and 20% HL-1 medium. The cell concentration was 10~5/ml. PHA was used to stimulate cell mitosis.
