Guanine nucleotide exchange factor (GEF) is essential for small G protein to switch on downstream signaling pathway,involved in cell adhesion and morphology.Rap GEF is a class of GEF and previous research has showed the role in Intergin and DE-cadherin mediated cell adhesion.One of Rap GEF,Gef26 may also impact the formation of synapse,a special cell adhesion,even nerve system development.Using NMJ of Drosophila as a model,we apply in situ hybridization,immunohistochemical staining,electrophysiology and western blot to explore the function of Gef26 in nerve development as well as its signaling pathway.The results show Gef26 is highly expressed in the central nervous system at the 16 stage of embryo.We observed significant reduction of NMJ boutons as well as branch length and decrease of transmitter release respectively in mutants of Gef26 via its target Rap1.Phosphorylation of MAPK (Erk),which relates to both of these two genes,increase in gef26 mutants.Together,these results demonstrate that Gef26 affects NMJ morphology and function by inhibiting Ras-MAPK function in MT organization.
