Current antidepressant paroxetine therapies have an unsatisfactorily high failure rate with up to 60% of depressed patients suffering from treatment-resistant depression (TRD).The aim of this study was to investigate the underlying mechanisms leading to poor success rate of paroxetine therapy.ABC transporters at the blood-brain barrier (BBB), such as P-gp and BCRP, may restrict the uptake of CNS drugs into the brain, thus contributing to the treatment failure.In the present studv, the role of P-gp/BCRP was assessed in mice pretreated with corresponding reversal agents alone or in combination.Adult male Swiss mice were pretreated with normal saline, verapamil(30 mg/kg, ip), CsA(50 mg/kg, iv) or pantoprazole(40 mg/kg, iv).After pretreatment, mice received an intravenous dose (1 mg/kg) of paroxetine via tail vein.
