Catechol-O-methyltransferase (COMT), one of the phase Ⅱ metabolizing enzymes, plays important roles in the metabolsim of endogenous and xenobiotic catechols.COMT is a metal ions-dependent intracellular enzyme that catalyzes the transfer of a methyl group from common methyl donor S-adenosyl-L-methionine (AdoMet or SAM) to one of the catecholic hydroxyls.In human body, COMT participates in the metabolism of many neurotransmitters and its inhibitors play a critical role in the management of central nervous system (CNS) disorders such as Parkinson's disease.Although several probe substrates for COMT have been reported, all of the currently used probes are non-fluorescent probes and their metabolites are often detected by liquid chromatography-tandem mass spectrometer (LC-MS/MS).
