H19通过干扰hnRNP U-actin复合物抑制RNA聚合酶Ⅱ介导的转录

  Background & Aims: H19 is one of the earliest identified and studied long noncoding RNAs.It is presumed that H 19 is essential for regulating development and disease conditions, and is associated with carcinogenesis of many type cancers.But the biological function and regulation mechanism of this conserved RNA, particular on transcription remain largely unknown.Methods: We performed RNA pulldown, RNA immunoprecipitation, and deletion mapping to identify the proteins that are associated with H19.Besides, we employ EU incorporation, immunoprecipitation and western blotting to investigate functional aspects of H19.Results: Our research indicated that H19 is bound to hnRNP U, and the association locates at 5 882nt region of H19.Moreover, H19 disturbs the combination between hnRNP U and actin, and then inhibits phosphorylation of RNA polymerase Ⅱ (Pol Ⅱ) C-terminal domain (CTD), consequently inhibits RNA Pol Ⅱ mediated transcription.We also showed that hnRNP U is essential for H19-mediated transcription repression.Conclusions: We demonstrate that H19 inhibits RNA Pol Ⅱ mediated transcription through disturbing hnRNP U-actin complex.These data suggest that H19 could regulate general transcription and exert more comprehensive effects on organisms.
