基于砷剂的功能纳米药物体系的构建及其肿瘤诊疗应用

  三氧化二砷(Arsenic Trioxide,ATO)已经被FDA 批准用于治疗急性早幼粒细胞白血病[1],然而其对于实体瘤的临床实验结果不理想。主要原因是ATO 在溶液中主要以亚砷酸根离子的形式存在,难以有效运输到肿瘤部位。相对于传统化疗药物,纳米药物运输系统具有诸多优点,如靶向病灶部位,控制药物释放,能够实现诊疗一体化等[2]。我们课题组应用纳米载体装载三氧化二砷在治疗实体瘤方面取得了较好效果[3-6]。大孔介孔二氧化硅(Large-pore mesoporous silica,LPMSN)作为纳米载体,具有低毒性,而且表面易被修饰[7],能够同时负载用于成像的磁性纳米颗粒以及用于治疗的砷剂和其它药物,从而实现多药负载联合治疗的纳米药物诊疗体系。我们设计了一种LPMSN 包裹的Fe3O4 纳米颗粒,并在LPMSN 孔道内采用共沉淀方法负载ATO 药物,从而实现于一种基于砷剂的诊疗纳米药物体系的构建。
