In this paper, a simple and rapid sample pre-treatment method, High Shear Dispersing Emulsifier(HSDE) extraction, had been developed for the content determination of flavonoids in bee pollen by high performance liquid chromatography(HPLC) and compared with ultrasonic extraction and immersion extraction.The results shown that HSDE had higher efficiency on breaking the cell wall and extracting flavonoid compounds of the pollen than other two methods.The optimized HSDE conditions were as follows: shearing time was 3 min, shearing Speed was 19000 r/min, ratio of liquor to material was 20∶1 (v/w).The wall-broken rate of bee pollen was higher than 98% with this treatment.The detection of pre-treated samples was carried out on HPLC coupled with a diode array detector (DAD).
